What causes a tyre to lose air slowly with no puncture?

Usually corrosion on the bead seat of an alloy rim, which stops the tyre sealing properly. It is common on older alloys and is fixed by cleaning and resealing the bead rather than replacing the tyre.

Do bigger rims affect ride quality?

Yes. A larger rim means a shorter tyre sidewall, which gives sharper steering but transmits far more impact from potholes — and makes both rim and tyre more vulnerable to damage on rough roads.
